Can this extract transmission fluid through the fill tube?

I would imagine it could if the tubing will fit down the fill tube and reach the bottom of the transmission pan. Keep in mind this is not an ideal way of changing transmission fluid as you still have a filter to replace and it will not remove the fluid from the torque converter.

can you use portable air compressor similar to those use to inflate a tire or you need something more powerful ?

Yes, it can work with portable air compressor similar to those use to inflate a tire. It only needs an airflow. It doesn't require too much pressure.

Can you do the reverse? Such as add fluid back into a differential or transmission?

No, it can't. According to the working principle, it should have the function of injecting liquid. But it will mix a lot of gas in the liquid. gas is useless for differential or transmission.

Can this work with gasoline and Diesel fuel ?

Theoretically, it can be used to extract all fluids. But its volume is only 500ml, you may have to repeatedly extract gasoline or Diesel fuel.
