Q: Can you test honey with it?

I would imagine you could. I would not contaminate it with water unless it is a distillate or distilled. Should work fine. My only thoughts...pyrethrums would kill off the whole hive so be sure you know what your testing for🙂

Q: What is the meaning of the two blue circles? Is one pesticide specific?

If there is white color in one or both middle wells, the test is positive. The sample may contain organophosphate or carbamate pesticides, toxic/heavy metals or naturally occurring acetylcholinesterase inhibitors. If there is a light to dark green/blue color in both middle wells/circles, the test is negative.

Q: Will this test an alcohol based extract (lemon zest) for pesticides remaining in the extract?

I don't see why not. But it is possible that the alcohol could interfere with the test. What are you looking for? The way this test looks for Organophosphates is if I remember correctly is by using Acetylcholine and I assume it is bonded somehow with that blue coloring agent. When Organophosphates inhibit Acetylcholine the blue color is released because organophosphates are anticholinergic. So a quick search of whether or not alcohol has an effect on Acetylcholine should give a strait forward answer.
