Yes, it comes with the hardener. You will find it when you take off the clear/white plastic lid.

Do I have to use fiberglass patch before applying the Bondo body filler? Or I can use it without the fiberglass patch on the car

Whether or not you use a fiberglass screen/cloth embedded in the Bondo depends on the job you are doing. I've seen people use this stuff a half inch thick or more without any fiber glass reinforcement. I'd definitely forgo the fiberglass with 1/4" or less. If the dent is on an old car fender that will flex some when you hit pot holes, and the dent is more than about 1/8" then fiberglass may help avoid cracking of the Bondo. Most important is drilling a grid of holes about 1/8" diameter and spaced about 3/4" apart over the area to be filled. You will push some Bondo through those holes to help lock the Bondo onto the metal. Of course surface preparation is also important to adhesion. Follow instructions carefully for that. If you do use fiberglass, be sure there is a layer of Bondo under it as well as on top of each layer of glass and work the Bondo into it well. I.e., the glass needs to be fully embedded in Bondo. Bondo does not work on things like a plastic bumper that naturally flexes (Bondo does not flex much). There are special products and procedures for repairing plastic front/rear covers.
