Q: What size hole on the boat would you need for this fit into in inches?

The vent is for a 5/8" hose I believe the hole in my boat was 5/8" as well , if your hole is bigger you could always add an escutcheon/ washer.

Q: does this vent have a one way valve built into it?

No it’s just open to surface fixture with a screened port hole! I was planning on putting that porous hard type filter in it , like on small engine intakes, to more or less keep in but out but also just relieve any built up fumes . Fuel Pressure!
